As the first step of detection, the sampling quality directly affects the results of a series of subsequent detections, and this step is usually lack of enough attention or even ignored. To ensure the quality of sampling, in addition to the standardization of sampling operations, the first thing we need to do is choosing a sampling swab which is in high quality and suitable material.
Sampling swab material used in COVID-19 PCR testing
The US CDC has mentioned in “Interim Guidelines for Collecting, Handling, and Testing Clinical Specimens from Persons for Coronavirus Disease 2019 (COVID-19)” that “Use only synthetic fiber swabs with plastic or wire shafts. Do not use calcium alginate swabs or swabs with wooden shafts, as they may contain substances that inactivate some viruses and inhibit PCR testing.”
Sampling swab materials
1. Medical cotton swab
Medical cotton swab is the most common wood-rod cotton swab. It is cheap and easy to obtain, but it may inhibit the PCR reaction, and the wooden rod is not easy to break, so it is not suitable for virus sampling.
2. Rayon swab
It is a swab with a polyester fiber or rayon tip and a plastic rod. Suitable for the collection of virological specimens, with no breaking point.
3. Nylon swab
It is made of nylon fiber by spray technology and is currently widely used for sampling of respiratory viruses such as COVID-19. Compared with the former two, it has high water absorption, high virus release rate and high virus stability.
